abstract | PURPOSE: To determine the quantity of uric acid in high accuracy, by decomposing and removing hydrogen peroxide existing in a biospecimen with a reducing agent, oxidizing and decomposing uric acid in the specimen with an enzyme uricase to generate hydrogen peroxide and determining the produced hydrogen peroxide. n CONSTITUTION: A biospecimen such as urine, blood, saliva and gastric juice is treated with a reducing agent such as sodium borohydride or sodium nitrite to effect the decomposition and removal of hydrogen peroxide existing in the specimen. After the above pretreatment, uric acid in the specimen is oxidized and decomposed with an enzyme uricase to generate hydrogen peroxide. The hydrogen peroxide produced by the reaction is determined by chemical titration such as titration with permanganate, iodic acid, etc., optical methods such as emission spectrometry, absorptiometry, fluorometry and laser nephelometry, physical methods such as polarography, etc., and the quantity of uric acid in the specimen is determined from the determined quantity of hydrogen peroxide. n COPYRIGHT: (C)1988,JPO&Japio |
