titleOfInvention Production of n-cyclohexanecarbonyl-d-alanine
abstract PURPOSE: To obtain the titled compound useful as a synthetic intermediate for pharmaceuticals, in high yield with extremely excellent operability, by reacting D-alanine with cyclohexane-carbonyl chloride in the presence of an alkali and a phase-transfer catalyst. n CONSTITUTION: D-alanine is dissolved in an aqueous solution of an alkali (e.g. sodium hydroxide), a phase transfer catalyst of quaternary ammonium-type (e.g. benzyl tributyl ammonium chloride) is added to the solution and the reaction liquid is cooled to ≤10°C, preferably about 5°C. Cyclohexanecarbonyl chloride and an aqueous solution of an alkali are dripped at the same time into the cooled reaction liquid to adjust the pH to 12W13.5 and to effect the reaction at room temperature to obtain the objective compound. The molar ratio of the cyclohexanecarbonyl chloride to D-alanine is slightly excess to 1 and the amount of the catalyst is ≥0.5wt.%, preferably 2W5wt.% based on the cyclohexanecarbonyl chloride. n COPYRIGHT: (C)1988,JPO&Japio
