abstract PURPOSE: To culture a deep-sea microorganism in an optimum environment, by aseptically collecting a microorganism under high pressure and pressing the microorganism into a culture vessel. n CONSTITUTION: A collecting apparatus 3 containing aseptically collected deep-sea microorganism and maintained under high pressure is attached to an attaching part 36 through introducing pipes 34, 35. A tank 12 is sterilized by energizing a heater 54 attached to a culture tank 2 and hot water in a bath 8 is sent through a pump 10 and a valve 58 to a groove 23 of the tank 2 to keep the tank 2 at a prescribed temperature. High-pressure valves 25, 29, 30 of the tank 2 are opened, a culture liquid in a reserve tank 4 is pressed into the tank 2 are opened, a culture liquid in a reserve tank 4 is pressed into the tank 2 with a booster pump 5, the valve 25 is closed, an inlet-side high-pressure valve 36 of the attaching part 36 is opened and the culture liquid is filled in the introducing pipe 35 to a level just before the outlet valve 3b of the apparatus 3. The pressures in the introducing pipe 35 and a stainless steel pipe 40 are equalized, the valves 26, 37, 3a opened to equalize the pressures in the introducing pipes 34, 35 and pipes 38, 40, etc., and a piston lever 3c is pulled to transfer the microorganism into the tank 2. n COPYRIGHT: (C)1988,JPO&Japio
