abstract | PURPOSE: To obtain a stable label antigen, by allowing an antigen having several antigen determinants to leave antigen determinants enough to hold antigen property thereof so that a label substance is bonded to the rest of the antigen determinants through a monoclonal antibody. n CONSTITUTION: First, a monoclonal antibody against an HBcAg is labelled with peroxidase by maleic imide method. This peroxidase-labelled HBc monoclonal antibody liquor mixed with various densities of HBcAg liquids to obtain a peroxidase labelled HBcAg. When measuring an IgM type Anti-HBc, first, an anti-human IgM rabbit IgG is immobilized on a microplate and serum to be inspected is added thereto. After washed, said peroxidase-labelled HBcAg liquid is added to the mixture and washed. Subsequently, a solution of H 2 O 2 and o-phenylene diamin and then, H 2 SO 4 are added to the mixture and after the stoppage of reaction, absorbance is measured at 492nm. n COPYRIGHT: (C)1988,JPO&Japio |
