abstract | PURPOSE: To make analysis with high accuracy by mixing the liposome which is conjugated with an antigen or antibody on the surface and is internally sealed with an electron carrier as well as sample and complement and determining the electron carrier flowing out of the broken liposome. n CONSTITUTION: A goat IgG antibody converted to thiol is added to the liquid suspension of the liposome sealed internally with 1-methoxy-5-methyl phenadium sulfate as the electron carrier. Such suspension is subjected to a centrifugal sepn. to remove the unreacted goat IgG after reaction and is further suspended in a gelatin-contg. liquid. This liquid suspension is mixed with the sample contg. a liposome diluting liquid, guinea pig complex and anti-goat IgG antibody and after reaction, a nitrotetrazolium blue and NADH soln. are added to the suspension to effect reaction; further the reaction is ceased by adding hydrochloric acid and surfactant thereto. The light absorbancy at 570nm of this reaction liquid increases with an increase in the quantity of the anti-goat IgG antibody. n COPYRIGHT: (C)1988,JPO&Japio |
