abstract |
PURPOSE: To enable quantitative determination of a physiologically active substance such as anti-inflammatory substance useful as an inflammation marker, etc., in high reliability, by activating blood coagulation factor to convert prekallikrein in plasma to kallikrein and measuring the quantity of produced kallikrein. n CONSTITUTION: Human plasma is diluted with physiological saline water, added with a kaolin suspension as an activation agent for blood coagulation factor XII and made to react in ice water bath for 20min to activate blood coagulated factor XII and convert prekallikrein in the plasma to kallikrein. The reaction system is added with an inhibitor such as trypsin inhibitor originated from lima bean to prevent the formation of excess kallikrein and then added with D-prolylphenylalanylarginyl-p-nitroanilide as a synthetic substrate. The quantity of the physiologically active substance can be determined by measuring the quantity of produced kallikrein by the measurement of the quantity of produced p-nitroaniline from the absorbance at 405nm wavelength. n COPYRIGHT: (C)1988,JPO&Japio |
