PURPOSE:To easily carry out quantitative determination of the titled substance useful as a marker for the diagnosis of diabetes, etc., by selectively removing sugars from a specimen, treating 1,5-anhydroglucitol in the specimen with pyranose oxidase, etc., in the presence of oxygen and measuring the quantity of produced H2O2. CONSTITUTION:A specimen (e.g. human serum) is treated with a strongly basic anion exchange resin, etc., to effect the selective removal of sugars from the specimen. 1,5-Anhydroglucitol in the treated specimen is made to react with pyranose oxidase or L-sorbose oxidase in the presence of oxygen and the quantity of produced hydrogen peroxide is measured to enable the quantitative determination of 1,5-anhydroglucitol. The measurement of hydrogen peroxide can be carried out by reacting the solution containing hydrogen peroxide using 2,2'-azinobis(3-ethylbenzothiazoline-6-sulfonic acid (ABTS) as a color developing agent and measuring the absorbance of the color-developed solution. |
