abstract NEW MATERIAL:An enzyme having the following properties. Catalytic activity, having catalytic activity to enzymatic reaction of formula 1 in the presence of Mg<2+> or Mn<2+> and free from catalytic activity to enzymatic reaction of formula 2; substrate specificity, capable of taking ammonia and NH4<+> as substrate and incapable of taking glutamine and asparagine as substrate; molecular weight, 50,000+ or -5,000; optimum pH, 8.5-10; pH stability, stable at pH 7.5-9 by the treatment at 60 deg.C for 15min; thermal stability, stable at <=60 deg.C by the treatment at pH 8 for 15min. USE:Reagent for the determination of ATP, deamido-NAD, ammonia or NH4<+>. PREPARATION:Bacillus stearothermophilus H-804 (FERM P-8839) which is a bacterial strain belonging to Bacillus genus and capable of producing the above enzyme is cultured preferably at 55-60 deg.C for 10-20hr and the objective enzyme is collected from the cultured product.
