abstract PURPOSE: To obtain a carrier having a large amount of ligand bonded thereto and free from adsorption by hydrophobic interaction, by immobilizing ligand on amino groups of a polymer-based carrier body subjected to an amination by an amino polymer or imino polymer employing a covalent bond through a crosslinking agent. n CONSTITUTION: Epoxidation is performed for the bonding of epichlorohydrin to -OH groups of a polymer-based carrier body to activate the surface of the carrier body. And then, the work is washed thoroughly by a buffer and subjected to an amination. After the suction of the supernatant of the solution, an aqueous solution containing 1,6 diaminohexane with the molecular weight smaller than that of the polyethylene imine is added to the work to perform an amination of the epoxy group that has survived the amination by the polyethylene imine. Thereafter, the gel is washed by highly pure water to complete the amination treatment of the surface of the carrier body. Subsequently, an antibody is immobilized on the surface of the carrier body thus subjected to the amination through glutaric aldehyde as crosslinking agent. n COPYRIGHT: (C)1988,JPO&Japio
