titleOfInvention Secretory protein induced by carcinogenic gene
abstract PURPOSE:To obtain a secretory protein induced by a carcinogenic gene and useful for the diagnosis and remedy of cancer, by specifying the molecular weight and isoelectric point of protein produced by culturing a cell originated from human hepatoma cell. CONSTITUTION:A human hepatoma cell (e.g. HuH-7cl-4) is inoculated in a medium in a Petri dish at a density of 0.5X10<4>-5X10<4> cells/cm<2> and cultured at about 37 deg.C for 1-4 days in an atmosphere having low oxygen partial pressure and a CO2 concentration of 3-10%. A plasmid (e.g. pSV2-myc) integrated with a carcinogenic gene [e.g. C-myc (a protooncogene corresponding to oncogene v-myc of chicken myelocytomatosis virus MC29)] is introduced into the obtained cultured cell. The introduced cell obtained by the above process is cloned and the separated cell is cultured in a medium such as Eagle MEM containing about 10wt.% bovine serum at a density of 10<4>-10<6> cells/cm<2> at about 37 deg.C for 2-4 days in an atmosphere containing 3-10wt.% CO2 to effect the secretion and production of protein in the medium. The objective protein having a molecular weight of about 46,000 and an isoelectric point of 7.1-7.3 can be separated from the cultured product after purification.
