titleOfInvention Measuring method of microorganism numbers
abstract PURPOSE: To measure microorganism numbers in a specimen in short time, by measuring fluorescence substance formed by reacting a nonfluorescence with an enzyme in a microorganism containing added cytoplasmic membrane- dissolving agent or lysosome and cytoplasmic membrane-dissolving agent. n CONSTITUTION: A solution or suspension which is a specimen for measuring numbers of microorganism is centrifuged and one or two or more kinds of 4MU derivative and AMC derivative are added to the resultant precipitation part. At the same time one or two or more kinds of cytoplasmic membrane- dissolving agents or lysosome and cytoplasmic membrane-dissolving agents are added to the precipitation. The cytoplasmic membrane-dissolving agent includes tributyltin, tripropyltin, etc. Then the mixture is kept at about 25W60°C for 30minW2hr and fluoroescence amount of AMC or 4MU produced in the reaction liquid is determined to measure numbers of microorganism in the specimen. n COPYRIGHT: (C)1988,JPO&Japio
