titleOfInvention Lectin-like protein originated from cultured cell, its production and antitumor agent composed mainly of said substance
abstract PURPOSE:To obtain the titled antitumor agent useful as a clinical or diagnostic drug for man, stably on an industrial scale, by culturing a cell originated from embryo of Sarcophaga peregrina (a kind of flesh fly) and purifying supernatant of the medium e.g. by ion-exchange chromatography, etc. CONSTITUTION:Established cells (NIHSape-4) originated from embryo of Sarcophaga peregrina is cultured and the obtained supernatant of the medium is subjected to adsorption, separation and fractionation by ion-exchange chromatography and hydrophobic chromatography to eliminate existing inhibitor and then subjected to adsorption, separation and fractionation by affinity chromatography using galactose, etc., as a carrier. The fractionation product is purified to obtain the objective antitumor agent composed of a lectin-like protein having a molecular weight of about 185,000 (by gel filtration), exhibiting hemagglutination activity, etc., and dividable into 4 subunits having a molecular weight of about 32,000 (by SDS gel electrophoresis) and 2 subunits having a molecular weight of about 30,000.
