abstract |
NEW MATERIAL:Anti-human colon carcinoma monoclonal antibody ACC-574 belonging to IgG1 class and reactive with colon carcinoma. USE:Reagent for pathologic diagnosis of colon carcinoma and a remedy for colon carcinoma. PREPARATION:For example, a fragment of human colon carcinoma membrane is mixed with a aluminum hydroxide, etc., and the mixture is administered to a BALB/c mouse. Spleen cell is collected from the immunized mouse, subjected to cell fusion with a mouse myeloma cell and cultured in HAT medium to obtain a hybridoma. The hybridoma is cloned e.g. by limited dilution analysis to obtain a hybridoma strain capable of producing anti-human colon carcinoma monoclonal antibody. The hybridoma is cultured in a medium or administered to a mouse to obtain ascites and the objective monoclonal antibody ACC-574 is separated from the cultured product or ascites. |
