abstract PURPOSE: To enable quantitative determination of various bile acids in a specimen in high accuracy in a short time, by using an internal standard substance obtained by the hydrolysis of a condensation product of free bile acid and an acidic amino acid ester. n CONSTITUTION: A specimen solution containing bile acids and an internal standard substance which is a hydrolysis product of a condensate of a free bile acid such as cholic acid and an acidic amino acid ester such as diethyl glutamate, etc., is supplied from a sampler 21 to a pretreatment apparatus 2. The bile acid-containing elute obtained by the retreatment is poured into a separation column 3 and the bile acids are separated into individual components and eluted. The elute is introduced into an immobilized enzyme column 5 together with an enzymatic reaction reagent solution 4 containing a substance capable of performing enzymatic reaction with bile acid, e.g. nicotinamide adenine dinucleotide. The immobilized enzyme column 5 contains immobilized bile acid dehydrogenase. The eluted liquid is introduced into a detector 6 and the quantity of NADH is determined to determine the objective bile acid. n COPYRIGHT: (C)1988,JPO&Japio
