titleOfInvention Production of optically active homophenylalanine and intermediate therefor
abstract PURPOSE: To obtain an optically active homophenylalanine useful as a raw material for ACE inhibitors in high purity and yield using an inexpensive resolving agent, by optically resolving a DL-homophenylalaline-optically active mandelic acid complex. n CONSTITUTION: DL-Homophenyalanine and optically active mandelic acid are dissolved in a solvent, e.g. acetic acid ester, etc., in the presence of an acidic compound, e.g. sulfuric acid or acetic acid, having preferably ≤5.0 pKa while heating at 20°C W boiling point of the solvent to provide a solution of two diastereomer salts, which is then cooled or concentrated to make the solution in a supersaturation state. A sparingly soluble diastereomer salt crystal, if necessary, is seeded to afford the optically active complex of the same kind. The pH of the solution is then adjusted to 4W6 using an alkali and crystals are filtered.
