abstract |
NEW MATERIAL:A monoclonal antibody reactive with natural and recombinant interferon-gamma and nonreactive with interferon-gamma decomposed by cyanogen bromide and/or pepsin. USE:A reagent for purification and quantitative determination of interferon-gamma. PREPARATION:A BALB/C mouse is administered and immunized with a non- glycosylated interferon-gamma. Spleen cell is extracted from the mouse and subjected to cell fusion with a mouse myeloma cell using polyethylene glycol to obtain a hybridoma. The hybridoma is screened to select a clone reactive with non- glycosylated interferon-gamma and cloned by limiting dilution analysis and the obtained monoclonized product is cultured to obtain the objective monoclonal antibody. |
