titleOfInvention Production of 1alpha-hydroxyvitamin d2
abstract PURPOSE: To readily obtain the titled compound in high yield, by passing through a novel intermediate ergosta-1,5,7,22-tetraen-3β-ol to provide 1α-hydroxy- ergosteryl diacetate and subjecting the resultant diacetate to irradiation with light, etc. n CONSTITUTION: Ergosta-1,4,6,22-tetra-3-ene expressed by formula I is reacted with isopropenyl acetate and the resultant product is subjected to reductive reaction to provide a novel intermediate expressed by formula II, which is reacted with 4-phenyl-1,2,4-triazoline-3,5-dione to afford a Diels-Alder type addition reaction product expressed by formula III. The resultant compound expressed by formula III is subjected to protection of OH group, epoxidation, deprotection and converted into a compound expressed by formula IV, which is then reduced and acetylated to give a compound expressed by formula V. The epoxy group is subsequently converted into ethylene group to afford a diacetate expressed by formula VI, which is then subjected to irradiation with light to provide 1α,3β-diacetoxyprevitamin D 2 . The obtained compound is further subjected to thermal isomerization and deacetylation reactions to give the aimed compound. n COPYRIGHT: (C)1988,JPO&Japio
