abstract | NEW MATERIAL:A compound expressed by formula I (X is H or, together with Y, forms methylene; Y is H, alkanoylthio or aroylthio when X is H; the broken lines at C 1 -C 2 and C 6 -C 7 are, as necessary, double bond). n EXAMPLE: 17-Hydroxy-15β, 16β-methylene-3-oxo-17α-pregna-4, 11-diene-21-carboxy lic acid.γ-lactone. n USE: A hypotensive and diuretic agent, diagnostic agent, remedy for aldosteronism, etc. n PREPARATION: For example, the 16α-position of suitably protected 17- ethylenedioxy-3β-hydroxyandrosta-5,11-diene is halogenated to give a compound expressed by formula II (R is acyl, etc.), which is then dehydrohalogenated to form a double bond. β-Methylene group is then introduced into the double bond, and further ethynyl group is introduced into the ketone at the 17-position to afford a compound expressed by formula III, which is then converted into a hydroxy.carboxylic acid. The resultant hydroxy.carboxylic acid is dehydrated and cyclized, and the OH group at the 3-position is oxidized to give the aimed compound expressed by formula I (X and Y are H; the broken lines are not double bond). n COPYRIGHT: (C)1987,JPO&Japio |
