PURPOSE: To obtain albumin in high purity and in high yield from a protein solution containing albumin derived from human plasma, by heat-treating the solution under a specific condition. n CONSTITUTION: A protein solution containing albumin derived from human plasma, especially from α- and β-globulin fraction is adjusted in 0.5W3W/V% protein concentration in the presence of 3W10mM organic acid having 3W10C or a salt thereof (e.g. butyric acid, caprylic acid, mandelic acid, etc.) and 1W10W/V% ammonium sulfate to pH 4.5W5.5, preferably pH 4.9W5.4, more preferably pH 5.0W5.3 and heat-treated at 65W70°C, preferably at 68°C for 15W60min, preferably 30min. After the heat treatment, the solution is cooled to 10W20°C, filtered, centrifuged (5,000W15,000rpm for 10W30min) and the supernatant liquid is recovered to give albumin. Consequently high-quality albumin preparation is obtained and this method can be applied to treatment of crude albumin fraction having undergone ammonium sulfate fraction treatment. n COPYRIGHT: (C)1987,JPO&Japio |
