abstract | PURPOSE: To enable the accurate and quick enzyme-labeling determination of trace substance useful in the fields of immunodiagnosis, biochemistry and fermentation industry, etc., by using a specific enzyme as a labeling substance. n CONSTITUTION: An enzyme (A) such as phosphoglucokinase is used as a labeling substance (enzyme) and bonded to an antigen, etc. The product is made to react with a substance (B) to be determined (antigen) and the reaction mixture is added with phosphoglucomutase (C), glucose 6-phosphate dehydrogenase (D), glucose 1-phosphate (E) and ATP and NAD(P)(F) to effect enzyme-A reaction. The component E is converted to glucose 6-phosphate with the enzyme C using glucose 1,6-2-phosphate (G) produced by the above reaction as a catalyst and, at the same time, the component F is converted to NAD(P)H(H) by conjugated reaction with the enzyme D.
