titleOfInvention Production of n-acetylneuraminic acid lyase
abstract PURPOSE:To produce a large amount of NA lyase at a low cost on an industrial scale, by introducing a vector integrated with a DNA carrying a genetic information concerning the production of N-acetylneuraminic acid lyase and culturing the microbial cell in a nutrient medium containing molasses, etc. CONSTITUTION:A vector integrated with a DNA carrying genetic information concerning the production of N-acetylneuraminic acid lyase is introduced in a microbial cell. The cell is cultured in a nutrient medium containing one or more kinds of components selected from molasses, glycerol and corn steep liquor to effect the accumulation of N-acetylneuraminic acid lyase in the medium. After the completion of culture, the microbial cells are collected from the cultured liquid e.g. by centrifugal separation and are disintegrated e.g. by ultrasonic treatment to extract an enzyme. The extracted liquid is treated e.g. by salting-out to obtain purified NA lyase. The above microbial strain is e.g. Escherichia coli H3-4.
