abstract | PURPOSE: To determine the antibody to the antigen A in human serum, by using a conjugate of normal human immunoglobulin and an antibody derived from an animal to the antigen A as a standard substance. n CONSTITUTION: In order to measure the anti-thyroxine antibody in human serum, an enzyme labelled thyroxine is prepared as a standard antigen (a), a 10mM phosphate buffer solution containing 0.5% gelatin, 0.1% bovine serum albumin and 0.3M NaCl as a buffer solution (b), a 0.1ml polystyrene column packed with anti-human IgG antibody insolubilizing cephalose as a column (c), 33mM o-nitrophenyl-β-D-galactoside solution as a substrate (d) and a 80mM sodium carbonate solution as a color former (e). 500μl of the labelled antigen (a) is added to 100μl of a diluted standard solution and 100μl of diluted serum to perform reaction for 2hr at 37°C. After the column (c) is washed with the buffer solution (b), the substrate (d) is made to flow through the column (c) and, after the column (c) is allowed to stand for 90min at 25°C, the absorbancy of the washing solution obtained by washing the column (c) with the color former (e) at 420nm is measured. n COPYRIGHT: (C)1987,JPO&Japio |
