abstract | PURPOSE: To realize culture of gene recombinant bacterial cell in high cell concentration without causing falling-off of plasmid and to enable mass production of the objective substance, by successively adding a culture-suppressing substance and an agent for preventing falling-off of plasmid to the culture medium. n CONSTITUTION: A gene recombinant cell (e.g. E.coli) retaining a composite DNA composed of the objective gene, a vector and a promoter in the cell and capable of expressing said objective gene is subjected to fed-batch culture. In the above culture process, the culture system is continuously or intermittently added with (A) a substance suppressing said promoter (e.g. tryptophan when the promoter is tryptophan promoter) and (B) an agent corresponding to the drug-resistant gene code to said composite DNA (e.g. ampicillin, kanamycin, etc.). n COPYRIGHT: (C)1987,JPO&Japio |
