titleOfInvention Lignin degradation enzyme and production thereof
abstract PURPOSE:To provide a lignin degradation enzyme obtained by the extraction and purification of cultured Lenzites betulina, requiring oxygen in enzymatic action and inactive to a compound corresponding to a lignin model compound wherein 4-site phenolic hydroxyl group is converted to methoxy group. CONSTITUTION:A lignin degradation enzyme which is an extracellular enzyme produced by culturing Lenzites betulina (a kind of wood decay fungi) in a medium. The enzyme contains copper, exhibits deep blue color as an aqueous solution, has an isoelectric point of about 3.5, requires oxygen to exhibit the enzymatic action, has an optimum temperature of about 50 deg.C (stable up to 50 deg.C) and optimum pH of 4.0-5.0 (stable pH range is 7.5-9.0), produces 2,6- dimethoxyphenol by acting to syringyl glycerol-beta-stringyl ether and is capable of depolymerizing lignin existing in the waste liquid discharged from the multi- stage bleaching step in the chemical pulp production process.
