abstract |
PURPOSE:To increase an amount of tryptophan collected from a culture mixture, by preparing a plasmid containing both a specific tryptophan operon and a gene conducting biosynthesis of serine hydroxymethyltransferase. CONSTITUTION:A plasmid containing both a tryptophan operon having released feed back inhibition of tryptophan on anthranilate synthase and a gene conducting biosynthesis of serine hydroxymethyltransferase is prepared. A bacterium belonging to the genus Escherichia, which is transformed with this plasmid and deficient in a gene conducting biosynthesis of tryptophan repressor and tryptophanase is cultivated in a nutritive medium and tryptophan is collected from the culture mixture. The nutritive medium preferably contains glycine. |
