abstract PURPOSE: To permit quick operation and specific response only to the fatty acid by fixing ≥5 kinds of immobilized enzymes such as thiokinase to an oxygen electrode. n CONSTITUTION: Five kinds of the enzymes; thiokinase, AMP deaminase, 5'- nucleotidase, nucleoside phospholilase, and xanthineoxidase are used as an enzyme film 2. This enzyme film 2 is disposed in the lower part of the oxygen electrode 10, is supported 3 on the outside thereof and is fixed to a vessel 11 by an O-ring 4 from the outside of a diaphragm 15 for the oxygen electrode. The fatty acid diffuses into the enzyme film 2 when a prescribed amt. of the standard sample of the free fatty acid is added into a buffer soln. Xanthine is finally formed and at the same time, the oxygen is consumed. As a result, the quantity of the oxygen diffused from the buffer soln. into the electrode decreases and therefore, the current value decreases. However, the fatty acid and oxygen diffusing from the buffer soln. into the enzyme film 2 are made stationary and the stationary current value is obtd. The quantity of the fatty acid in the buffer soln. is, therefore, measured from the current value. n COPYRIGHT: (C)1987,JPO&Japio
