abstract PURPOSE: To obtain the titled factor giving little grave side effects and selectively acting to tumor cell, by extracting disintegrated platelet with acidic aqueous solution or mixed water and an organic solvent solution and fractionating the extract by gel-filtration and chromatography. n CONSTITUTION: Platelet subjected to cytoclasis by freezing and thawing or with a homogenizer is extracted with an aqueous solution of an inorganic or organic acid or a solution of the acid in a mixture of water and an organic solvent (preferably 1M acetic acid or 75% ethanol containing 1M acetic acid) and, if necessary, subjected to gel-filtration to obtain a fraction having an estimated molecular weight of 50,000W150,000 and effective in suppressing the proliferation of tumor cell. The fraction is subjected to ion-exchange chromatography, reverse-phase chromatography, etc., to form a fraction active to suppress the proliferation of tumor cell and having the following properties. Molecular weight, 50,000W150,000; isoelectric point, 4.5W5.3; ninhydrin reaction, positive; stability, deactivated with trypsin and stable by heating at 60°C for 30min in physiological saline water or contacting with 0.2N hydrochloric acid at room temperature for 60min or 70% formic acid for 30min. n COPYRIGHT: (C)1987,JPO&Japio
