abstract | PURPOSE:To impart hydrogen productivity 2-3 times as high as that of ordinary plasmid, by integrating a hydrogenase based gene present on chromosomic DNA of a microorganism of the genus Citrobacter and forming a hybrid plasmid. CONSTITUTION:A hydrogenase based gene present on chromosomic DNA of a microorganism of the genus Citrobacter, e.g. Citrobacter freundii, etc., is integrated to form a hybrid plasmid. The operation to extract the chromosomic DNA from bacterial cells can be performed by, e.g. Saito-Miura et al. method. The operation to fragment the extracted chromosomic DNA can be carried out by a restriction enzyme or endonuclease having no base sequence specificity. Examples of a microorganism receiving the hybrid plasmid containing integrated hydrogenase based gene to be used include microorganisms of the genuses Escherichia and Citrobacter. pCBH2 having 6.4 Md molecular weight and pCBH4 having 3.7 Md molecular weight are cited as the above-mentioned plasmid and restriction maps are shown in the figures. |
