abstract PURPOSE:To produce a plasmid and microorganism by including a DNA region coding the amino group terminal of a metapyrocatechase gene and the region coding the peptide corresponding the amino acid numbers of from 92 to 105. CONSTITUTION:A bacterial strain belonging to Escherichia genus is transformed by a plasmid containing a high-expression promoter operon, the DNA region coding the amino group terminal of the metapyrocatechase gene containing the SD sequence of the metapyrocatechase gene positioned in the downstream region subjected to the above operator, the region connected to the above DNA region and coding the peptide corresponding at least the amino acid numbers of from 92 to 105 of the sabin 1, sabin 2 or sabin 3-type capsid protein VPI of attenuated poliovirus strain, and the region coding the ampicillin resistance and positioned in the upstream side of the above region. The transformed strain is cultured in a nutrient medium, and the fused protein accumulated in the bacterial cell is separated therefrom.
