abstract | PURPOSE:To perform quantitative analysis simply with high sensitiviy, by performing the quantitative analysis of inosine by immunoassay. CONSTITUTION:In this quantitative analysis of inosine, acid anhydride and org. tertiary amine are at first added to a specimen and a standard solution to perform the acylation of the hydroxyl groups at the 2'- and 3'-positions of inosine. After the above-mentioned acylating reaction, a measuring specimen is diluted by 5-10 times in usual by a buffer solution and an inosine standard solution is successively subjected to multiple dilution by a diluting buffer solution and both diluted solutions are subjected to antigen-antibody reaction. The antigen-antibody reaction is performed by adding 2',3'-diacylinosine in the measuring specimen and a fixed amount of labelled 2',3'-diacylinosine to a fixed amount of a soluble or insoluble anti-inosine antibody simultaneously or almost simultaneously. After the completion of the antigen-antibody reaction, labelled 2',3'-diacylinosine bonded to the antibody and that in an unbonded liberated state are separated by a usual method and the labelling amount of either one of them is measured. |
