abstract PURPOSE:To provide high detecting sensitivity and to make quick measurement even with a bodily fluid such as whole blood contg. the macromolecules such as blood cells by using cloth for a developing layer of a multi-layered analyzing element for quantitative determination of cholesterol and incorporating a reagent for detecting cholesterol therein. CONSTITUTION:A water absorbent layer 12 consisting of a hydrophilic high polymer and further cloth incorporated therein with the reagent for detecting cholesterol thereon are successively laminated on a water impermeable and light transmittable base 10. When a simple liquid is spotted to the layer 14, the ester type cholesterol in the sample liquid is hydrolyzed in the layer 14 and forms the free type cholesterol. H2O2 is generated therefrom by a cholesterol oxidase action agent. The H2O2 and a coloring reagent compsn. form a colored material by the peroxidase effect and said material is diffused and migrated into the layer 12. The total content of the cholesterol in the sample liquid is thus quantitatively determined from the quantity of the colored material by measuring the optical density of the reflected light through the base 10. Since the pulverous particles which shield the color are dispersed and incorporated into the layer 14, the measurement of the optical density of the reflected light is not disturbed even if the colored sample such as whole blood is used.
