abstract | PURPOSE: To enable the formation of several kinds of enzymes at a part of a solid surface as immobilized enzyme film, by arranging a process of forming an enzyme film, a process of performing a crosslinking reaction and a process of removing an enzyme film. n CONSTITUTION: A protein solution containing a first enzyme is used to form a first enzyme film 3 on the surface of a wafer in which an island-like silicon layer on the surface of a sapphire substrate 1 is used to form a sensor section 2 of an ISFET. Then, the wafer surface is covered with a metal mask 5 and a crosslinking agent solution 4 is sprayed onto the metal mask 5. Then, glutaric aldehyde solution drops are made to reach the enzyme film 3 adhered on the sensor section 2 through an opening of the metal mask 5 provided so as to be positioned on the sensor section to perform a crosslinking reaction with the enzume film 3. Then, after allowed to stand at the room temperature for about 10min., the work is washed sequentially to remove the unreacted glutaric aldehyde and all the enzyme film 3 but that immobilized on the specified sensor section 2.
