abstract | PURPOSE:L-Phenylalanine dehydrogenase useful for producing L-phenylalanine, by cultivating a specific bacterium belonging to the genus Sporosarcina. CONSTITUTION:0.01-10W/V% L-phenylalanine as an induction substance is added to a base medium containing a nitrogen source (e.g., peptone), a carbon source (e.g., starch), etc. to give a medium. Sporosarcina ureae is inoculated into the medium and cultivated aerobically at 25-45 deg.C at 6-11pH or 6-48hr. A mold is collected from the culture mixture and it is ground to give a crude enzyme. Then it is subjected chromatography, treated with polyethylene glycol and crystallized to give an enzyme having the following properties wherein it catalyzes a reversible reaction shown by the formula (amount of each compo nent is 1mol), shows about 290,000 molecular weight by high performance liquid chromatography gel filtration method and a subunit having about 38,000-39,000 molecular weight by SDS-polyacrylamide gel disc electrophoresis method and acts specifically on L-phenylalanine of the table (oxidative deamination reaction). |
