titleOfInvention Production of 1,4-dihydropyridine derivative
abstract PURPOSE: To obtain the titled compound having pharmaceutical actions such as blood flow increasing action, vasodilating action, antihypertensive action, etc. by a simple operation in high yield, by reacting a novel α-benzyl-β-keto-ester derivative with an enaminocarboxylic acid derivative. n CONSTITUTION: A β-keto-ester derivative is reacted with a corresponding ben zaldehyde compound in the presence of a halide to give a novel α-benzyl-β-keto- ester derivative shown by the formula I (R 1 and R 2 are H, halogen, nitro, or CF 3 ; with the proviso that R 1 =R 2 ≠H, and R 3 and R 4 are lower alkyl; X is halogen) easily in high yield. This compound is reacted with an enamino carboxylic acid derivative shown by the formula II [R 6 is as shown for R 3 and R 4 ; R 5 is R 6 , or (substituted)aralkyl; n is 1W3] preferably in a solvent at 30W120°C optionally in the presence of a base for 30minW24hr, and, then option ally subjected to a salt formation reaction, to give the aimed compound shown by the formula III to be expected as an improved agent for circulatory organs or its nonpoisonous salt. n COPYRIGHT: (C)1986,JPO&Japio
