abstract |
PURPOSE: Cow milk is passed through an affinity column filled with insoluble support to which monoclonal antibovine lactoferrin antibody is immobilized to enable high-yield separation and high-purity purification of the titled substance which binds to iron, thus having bacteriostatic action on iron-requiring bacteria. n CONSTITUTION: Mouse spleen lymphocytes immunized with bovine lactoferrin are fused with mouse myeloma cells and the resultant hybridoma is cultured to produce monoclonal antibovine lactoferrin antibody. The resultant antibody is immobilized on a support for affinity chromatography by stirring them overnight. Cow milk is passed through the column to effect adsorption of bovine lactoferrin, which is then eluted with an eluent of a less than 4.7 pH, preferably 4.3W2.7 pH, then the eluted fraction is immediately neutralized to effect isolation and purification of the objective substance. n COPYRIGHT: (C)1986,JPO&Japio |
