NEW MATERIAL:A monoclonal antibody which is cross-reactive only to one of antigen-determining groups in rat prolyl hydroxylase. n USE: A reagent to determine prolylhydroxylase in blood whereby the fibrillation of hepatic tissue is judged. n PREPARATION: Skins of rat fetuses are extracted, fractionated with ammonium sulfate, then subjected to a column to which poly-L-proline is immobilized to collect and purify rat prolylhydroxylase. The enzyme is given together with complete Freunds's adjuvant intraperitoneally to Balb/C mouse to effect immuni zation. Then, spleen is removed to collect its cells. They are fused with myeloma cells and cultured in the HAT medium to collect only hybridoma. The hybriodomas are screened and monocloned by the limiting dilution, then the monocloned cells are cultured to obtain the objective monoclonal antibody. n COPYRIGHT: (C)1986,JPO&Japio |
