titleOfInvention Colorimetric measuring method of ceruloplasmin
abstract PURPOSE:To provide a colorimetric measurement method which can make measurement with a small amt. of specimen in a shorter period, permits easy operation, has the excellent stability and reproducibility of sensitivity and is suitable for an automatic analyzer by using a 4-aminopyrazolone deriv. and aniline, etc. as a color forming agent. CONSTITUTION:The 4-aminopyrazolone deriv. and aniline or the deriv. thereof or the salt thereof are used. A buffer soln contg. either phenol or phenol deriv. is used. A buffer soln. contg. a benzothiazolinone hydrazone deriv. or the salt thereof and aniline or the deriv. thereof or the salt thereof is used. The specimen is added to these solns., by which the coloration reaction is initiated. The amt. of the reactive liquid and the specimen is suitably 2X3ml reactive liquid with respect 20mul specimen. The reaction temp. is suitably about 37 deg.C. On the other hand, the coloration sensitivity is measured by making direct absorptiometric measurement of the quantity of the dye formed after lapse of the suitable specified time, by which the measurement of the activity of ceruloplasmin is made possible as well.
