abstract | PURPOSE:To obtain a ninhydrin reagent for quantitative analysis of amino acid having no danger generating a noxious substance inducing blood disorder or teratogenesis as a byproduct, by using specific 1-alkoxy-2-alkanol alone or in combination with dimethylsulfoxide. CONSTITUTION:1-Alkoxy-2-alkanol represented by formula (wherein R<1> and R<2> are respectively independently 1-4C lower alkyl) is used alone or in combina tion with dimethylsulfoxide to be used as the solvent of a ninhydrin reagent for quantitative analysis of amino acid. This solvent eliminate such a problem that methyl cellosolve conventionally used as a solvent generates methoxyacetic acid being a side effect substance bringing about blood disorder or atrophy of a spermary. This reagent can be used without showing no lowering in colora tion over ten days and more and has good sensitivity and is prevented from the precipitation of a crystal during the preservation in a refrigerator and can be safely used without generating the clogging of the line in an automatic analyser. |
