titleOfInvention Reagent for quantitative determination of theophylline
abstract PURPOSE:To measure the theophylline in a bio-fluid with high sensitivity and high accuracy without the need for a special pretreatment of the bio-fluid by using the antibody formed by injecting the conjugate of a specific theophylline deriv. and protein to an animal and the deriv. labelled by enzyme, etc. CONSTITUTION:The theophylline which is expressed by the formula [R is H, -(CH2)nR', n is 1-5, R' is -COOH, -CHO or -NH2] is modified in the 8th position by S to form the theophilline deriv. Such deriv, is conjugated with protein such as albumin of a cow serum, etc. and the resultant conjugate is injected to an animal and the antitheophylline antibody is obtd. from the antitheophylline serum of said animal. On the other hand, the labelling antigen formed by labelling the compd. expressed by the formula with beta-D-galactosidase is prepd. The dilute soln. of the bio-fluid (human serum, etc.) as well as the above-described antitheophylline antibody and the labelling antigen are brought into competitive reaction and thereafter the substrate is brought into reaction. The absorbancy of the color formed by the reaction is measured and the measurement of the theophylline in the specimen from the preliminarily obtd. standard curve is made possible with high sensitivity and high accuracy.
