abstract PURPOSE:To enable early diagnosis of a tuberculosis patient by bringing the sensitized red blood cells obtd. by conjugating a tubercle bacillus phospholipid antibody via a coupling agent to immobilized red blood cells and a human bodily fluid or the dilute liquid thereof into contact with each other, observing the agglutinated image thereof and detecting the tubercle bacillus phospholipid with high accuracy. CONSTITUTION:The live red blood cells obtd. from a goat, rabbit, etc. is subjected to a gaseous CO treatment and formalin is added thereto to obtain the immobilized red blood cells. The tubercle bacillus phospholipid antibody is conjugated via the coupling agent such as tannic acid to such red blood cells to obtain the sensitized red blood cells. The sensitized blood cells are stable for >=1 year if the blood cells are suspended in a physiological salt soln. contg. PBS and BSA added with sodium azide and are preserved in a frozen chamber. The sensitized blood cell suspension is brought into contact with a sample serum or the dilute liquid thereof and the agglutinated image thereof is observed. The agglutinated image of the double-double dilute liquid of the sample contg. standard phospholipid is compared with the agglutinated image of the sample and the measurement of the concn. of the phospholipid in the same with good accuracy by using a slight amt. of the sample is made possible.
