titleOfInvention Method and reagent for quantitative determination for automatic analysis of ammonia in blood
abstract PURPOSE:To enable quantitative determination of ammonia in blood by various kinds of automatic analyzing instruments by using specific reagents. CONSTITUTION:The buffer soln. such as a phosphoric acid buffer soln., Good buffer soln. or tris buffer soln. or the like contg. 1-10mM alpha-ketoglutalic acid and 0.5X10<-4>-1.0X10<-4>M NADP Hz and having 7.0-8.5pH is used as the 1st reagent and the similar buffer soln. contg. 30-60% (V/V) glycerol and 0.2- 0.5mM disodium ethylenediaminetetraacetyl (EDTA.2Na), having the activity value of NADP Hz dependency type GLDH adjusted to 5IU/ml and having 6.0- 6.5pH is used as the 2nd reagent in this measuring method. The above-mentioned 1st and 2nd reagents are successively added to the specimen serum and the concn. of ammonia in the blood is determined from the difference between the absorbancy of the characteristic absorption of the NADP Hz measured after addition of the 1st reagent and the absorbancy of the characteristic absorption of the NADP Hz measured after the end of the reaction reduced by addition of the 2nd reagent.
