abstract PURPOSE: To enable the quick and convenient detection of the inhibiting substance of a starch hydrolase, by applying a thin starch layer directly on a flat gel plate, and detecting the influence of the inhibiting substance on the starch diffused in the gel directly by iodine reaction. n CONSTITUTION: Agar, etc, is poured into a flat vessel and gelatinized. A thin film of starch is placed on the above thin gel layer and left at room temperature to effect the diffusion of the starch into the gel, and the surface of the gel is sprinkled lightly with running water to remove the starch film. Specimen cavities are bored to the gel at proper intervals, and are charged with the solution of the test specimen. The solution in the cavity is left to stand until the water in the cavity is almost absorbed in the surrounding gel, when a gel containing starch hydrolase is placed on the gel to effect the diffusion of the enzyme into the base gel. A solution of iodine is applied to the surface of the base gel, and after a little while, the vessel is dipped in running water to remove the color. A clear pigment ring can be discerned around the specimen cavity containing the enzyme inhibiting substance. The substance can be detected thereby. n COPYRIGHT: (C)1985,JPO&Japio
