titleOfInvention Determination of 3alpha-hydroxysteroid
abstract PURPOSE: 3α-Hydroxysteroid (bile acid) in the sample to be tested is converted into nicotin-adenine nucleotide of reduced type (NADH), then a metal ion is allowed to act on the NADH and the amount of the metal complex formed is determined, thus even a trace amount of sample has become determinable. n CONSTITUTION: 3α-Hydroxysteroid dehydrogenase (3α-HSD) is allowed to act on 3α-hydroxysteroid in the sample to be determined in the presence of nicotin- adenine nucleotide (NAD) to form NADH. Then, the electrons of NADH are allowed to act on metal ion using an electron carrier such as 1-methoxy-5-methylphenazinium methyl sulfate in the presence of an organic acid salt oxamate or oxalate to form a reduced metal ion and a specific metal color reaction reagent is allowed to act on the ion to form the metal complex and the complex is determined. n COPYRIGHT: (C)1985,JPO&Japio
