titleOfInvention Production of antiimmune complex antibody
abstract PURPOSE:To enable exact quantitative determination of an antiimmune complex by using a soln. prepd. by producing and drawing the antibody for other animals, etc. with the agglutinated matter of F(ab')2 fragment of human immune globulin as an antigen and refining the same by dialysis, etc. or the freeze dried matter thereof. CONSTITUTION:Rabbit, etc. are immunized and are let with blood by using, as an antigen, the F(ab')2 fragment prepd. by subjecting the serum, etc. separated from a human blood, etc. for blood transfusion to ammonium sulfate salting out and further fractionating the same with a DEAE cell roll ion exchange chromatography to obtain IgG, etc., and refining the same by pepsin decomposition, etc. Ammonium sulfate is added to such serum and a precipitate is drawn, which is dialyzed with a buffer soln. of 8pH and the fraction passing through a DE-52 column is further refined by a dialysis treatment, etc. to obtain an antibody. The antibody obtd. by freeze drying such antibody or separating the mouth spleen cells obtd. by injecting F(ab')2 thereto, bringing the same into plasmoganny with mouse myeloma P301 and culturing the antibody is used. The specific detection of the immune complex, etc. such as human IgG or the like is thus made possible.
