titleOfInvention | Preparation of immobilized enzyme |
abstract | PURPOSE: To prepare an immobilized enzyme capable of keeping high enzymatic activity for a long period, by saponifying a specific water-dispersible polymer particle, and immobilizing an enzyme through the produced hydroxyl group with a covalent bond. n CONSTITUTION: Water-dispersible polymer particles having an average particle diameter of 0.03W2μ and containing ester group are prepared by the emulsion polymerization of (a) 30W98wt% of vinyl acetate and/or vinyl propionate, (b) 0W68wt% of a monomer copolymerizable therewith (e.g. styrene), (c) 1W20wt% of a polyfunctional monomer for internal crosslinking (e.g. ethylene glycol dimethacrylate) and (d) 1W60wt% of (meth)acrylonitrile, in an aqueous medium. The polymer is saponified with a 0.01W5N aqueous solution of an acid or alkali at 20W70°C to convert the ester group to hydroxyl group. The hydroxyl group is epoxidized and made to react with the amino group of an enzyme (e.g. catalase) to immobilize the enzyme to the polymer particle with a covalent bond. n COPYRIGHT: (C)1984,JPO&Japio |
