abstract PURPOSE: To prepare a novel thermostable alkali metal protease, by extracting the fruiting body of Grifola frondosa or its cultured body. n CONSTITUTION: The fruiting body of Grifola frondosa, belonging to Grifola genus, Polyporaceae family, or its cultured body is extracted with a salt solution having a pH of 3W10 and salt concentration of 0.05W0.5M preferably at ≤4°C, and the obtained extract liquid is purified to obtain the objective thermostable alkali metal protease having the following characteristics: Molecular weight, about 22,000; contains one Zn atom per one molecule of protein; optimum pH for the decomposition of casein, albumin, etc., 9W10; exhibits endopeptidase activity specifically hydrolyzing the peptide bond at the N-terminal of leucine, phenylalanine, valine and lysine residue. n COPYRIGHT: (C)1984,JPO&Japio
