abstract | PURPOSE: To determine the enzymatic activity, by reacting the substrate with the enzyme to be determined to produce the oxidizable coloring compound, then reacting the compound with an enzyme capable of converting the compound to a pigment in the presence of oxygen, and determining the produced pigment. n CONSTITUTION: A substrate is made to react with a specimen having the activity of an enzyme (Y) capable of converting the substrate to the oxidizable coloring compound (A) or an oxidizable condensed coloring compound (B) to obtain the compound (A) or compound (B). The compound (A) or (B) is treated with an enzyme capable of converting the compound to a pigment in the presence of oxygen and if necessary a compound producing a pigment when condensed with the compound (B). The activity of the enzyme Y in the specimen can be determined by determining the produced pigment. The enzyme Y is e.g. leucine aminopeptidase (LAP), γ-glutamyl-transpeptidase (γ-GTP), etc., and the substrate is L-leucine-p-N,N-diethylaminoanilide for LAP and γ-glutamyl-3-carboxy-4- hydroxyanilide for γ-GTP. n COPYRIGHT: (C)1984,JPO&Japio |
