abstract PURPOSE: To determine NH 3 (or ATP) in a liquid specimen, in high sensitivity and accuracy, by utilizing the cycling reaction combining specific ensymatic systems. n CONSTITUTION: A specimen liquid containng NH 3 is added with (A) ATP, a water-soluble carbonate (e.g. NaHCO 3 ), carbamate Kinase and a water-soluble salt capable of releasing Mg 2+ (e.g. MgCl 2 ) and (B) a reagent containing the second kinase different from the above (e.g. pyruvate kinase) and a substrate phosphorus compound (e.g. phosphoenolpyruvic acid). The ATP is converted to ADP with the carbamate kinase, and at the same time, the formed ADP is converted to ATP and the substrate phosphorus compound is converted to a kinase reaction product (e.g. pyruvic acid) by the second kinase by this process. The quantity of the reaction product is determined to determine the quantity of NH 3 in the specimen liquid.
