titleOfInvention Rheumatic specific protein and antibody thereof
abstract PURPOSE:To obtain an antibody which does not exhibit false positiveness at all to an ordinary health person, and a renal insufficient or cancer patient at all by using the antibody obtd. by immunizing the specific protein which is obtd. by separating and refining by a specific method from rheumatic plasma and has a specific mol.wt., and a specific isoelectric point and mobility in an animal. CONSTITUTION:Ammonium sulfate is further added up to 35% satn. to the supernatant removed after a centrifugal sepn. of the precipitate of 25% satd. ammonium sulfate of the waste plasma formed by plasma exchange of a rheumatic patient and the resulted precipitate is centrifugally separated. The precipitate is dissolved in a phosphoric acid buffer soln. and the soln. is refined with dialysis, ion exchange chromatography, etc., by which the specific protein is obtd. This rheumatic specific protein exhibits about 170,000mol.wt. when measured with SDS polyacrylamide gel electrophoresis, has 7.3-7.8 isoelectric point in the secondary electrophoresis using no protein modifying agent and has the mobility in a range of 0.40-0.55 (with the mobility at the foremost of alubmin as 1.0). The simple and exact diagnosis of a rheumatic disease is made possible with high sensitivity by using the antibody obtd. by refining from the antiserum obtd. by a hypodermic injection of such protein to a house rabbit.
