titleOfInvention Preparation of specific antigen of human pregnancy
abstract PURPOSE: To prepare the titled antigen useful as an immunological contraceptive, etc., in high quality, with simple procedure, by using a material containing specific antigen of pregnant woman, e.g. placenta as a raw material, and subjecting the material to acid treatment, ammonium sulfate fractionation, gel-filtration treatment, and affinity chromatography. n CONSTITUTION: A liquid containing the specific antigen of pregnant woman (SP 1 ) (e.g. extract of human placenta, urine, blood, etc. of pregnant woman, etc.) is adjusted to 4W15pH, and the precipitate is removed. The residue is added with 3W10% saturation of ammonium sulfate, and the produced precipitate is removed. Additional ammonium sulfate is added to 50W70% saturation, and the precipitate is recovered. The precipitate is dissolved in a buffer solution having low ionic strength, and subjected to the gel-filtration fractionation with a carrier corresponding to a substance having a molecular weight of 50,000W500,000 Dalton to obtain a fraction having a molecular weight of 80,000W120,000. The obtained fraction is subjected to the affinity chromatography with a carrier having an anti-nonpregnant normal human plasma antibody as an affinity group to obtain the objective specific antibody for pregnancy. n COPYRIGHT: (C)1984,JPO&Japio
